misfire on No3 cylinder 400/4

Hi i have completely stripped my carbs on my 400/4 and replaced all the internals and cleaned thoroughly,they are now on the bike and set up , the bike fires up ok and runs but no 3 cylinder is not firing properly,i have checked the timing and points , compression and valves all seem ok but have run out of ideas now, has anyone got any ideas i can try thanks  Tony.

check for spark with a spare plug in the #3 wire. check that plug wire to see if its screwed into copper wire, or just rubber insulation.
