I'm rebuilding my cb 750K (1978) engine.  I've got the cylinders back on and in the process of putting the head back on, one of the bolts wouldn't tighten.  The threads are stripped that it goes in to.  I really don't want to pull the head off again (I'm concerned about the new gasket) to tap it.  Also, I've never tapped aluminum before and don't know how that will go.  So here's my questions:  1) how big a deal will it be to leave that bolt out and simply torque the rest of them down good? - or; 2) can I tab it without removing the head?

Not sure of your thread size, but a thread insert (Heli-Coil) could be done, even possibly without having to drill it out if it is stripped pretty bad already. Then you wouldnt have to worry about drilling a straight hole because you dont want to take the head off. You can get them at McMaster or I saw some at Advance Auto parts the other day...Hopefully you are not referring to the main studs that run down to the case, then it looks like it all needs to come back off to fix it.

no way man, take off the head for the love of god. go to your hardware store with one of your head bolts and get a helicoil set. comes with a tap, a helicoil driver and several helicoils for if you #$%* up the rest. try reusing your new head gasket, waddya got to loose? motor probably hasnt been started since the head was taken off, so it should be fine. if it runs bad do a compression test and maybe a combustion leak test (might wanna have a shop do that, don't pay too much, just tell em you think youve got a bad head gasket. but really a new headgasket can't be that much. might wanna tap and die the rest of your hardware so it doesnt happen again.

Which bolt? Pictures will help..

a big deal. good way to bend your head, and wear out the cylinders that ARENT leaking
