Can a back wheel be trued without removing from bike?

Just wondering..... I removed the front wheel because the bars turn messing up any precise gauge reading but the back wheel I just wanted a second opinion on since it stays put. Thank you in advance I really appreciate it.

Sure, why not?! Might not be QUITE as good or easy but you may get it spot on. I'd remove the chain and make sure it will  free wheel, set up your dial indicator and go for it.
As of today 3/13/2012 my original owner 75 CB750F has made it through 3 wives, er EX-wives. Free at last.  ;-)

Doesn't putting the bike in neutral free the wheel so that I don't have to remove chain?

I think he means so there is almost no drag. Then you can give it a good spin when youre done to see how straight it goes.
